Check the following factors for bud drop 1. Low light, 2. Not enough fertilizer 3. excessive heat and I will add the 4th factor overwatering? Datura wilts very easily in Karachi's heat but it dislikes over-watering. See if the soil is kept moist but not wet.

About Datura, is it the same one growing wild on roadsides etc? .. Given the leaves, and its flower fruit I think it is the same as "Dhatura" in the local language, which is quite a deadly poison

Farooq sb. then there could only be one reason. About 1st reason datura is getting about 8 to 9 hours of sunlight. For 2nd reason I added manure to it previously so it can not be reason as there in enough nutrients. For 3rd reason I watering it excessively which is definitely the reason behind. JazakAllah

How much is the toxicity. Because I have never seen any type of insect on it or near it. I have read about it and came to conclusion that the it is beneficial for asthma patients and Applying it on wound will help recover it.

Mealy bugs, beetles, spider mites.. all can attack it... it is toxic to higher animals.. causes hallucinations, difficulty in breathing and in higher doses, death... most toxic part are the seeds... the datura you have produces large fragrant flowers like these:
